Abstract: To determine the genetic relatedness of selected lines of carnation, two types of molecular markers, random amplified polymorphic DNA (RAPD) and inter-simple sequence repeat (ISSR) were assayed. A mean polymorphism information content value of 0.492 (RAPD) and 0.530 (ISSR) was found with both markers respectively. In RAPD analyses, 12 out of 28 bands (42.85%) were polymorphic unlike in ISSR analyses, a total of 96 bands were detected, among which 51 bands (53.12 %) were polymorphic. The RAPD-based genetic similarity (RAPD-GS) ranged from 0.66 to 1.0, with the mean of 0.845. The number of bands per primer is 5.6 using RAPD with a polymorphic range of 62.5 per RAPD primer pair, whereas using ISSR it is 19.2 number of bands per primer with a polymorphic range of 57.14 per ISSR primer pair. The ISSR-derived genetic similarity (ISSR-GS) ranged from 0.285 to 1.0, with the mean of 0.754. ISSR was better than RAPD to detect genetic diversity among the carnation lines. Cluster analyses indicated that both RAPD and ISSR markers could distinguish the carnation lines.
